چکیده

Chylomicron remnants can penetrate into the artery wall, where they can initiate atherogenesis. Since it is difficult to isolate these particles from human blood because of contamination with other lipoproteins, the use of lipid emulsions as chylomicron remnant-like particles (CRLP) has been proposed to study their metabolism. The present work was aimed to evaluate the methodology for the preparation of CRLP. Artificial chylomicrons were prepared by sonication of a lipid mixture and separated by density gradient centrifugation. Lipid classes were analyzed by HPLC and fatty acids by GC. Particle size was measured by dynamic light-scattering and the presence of apolipoprotein E by immunoblotting. The highest lipid content was found in the 60400. This latter fraction presented the highest triacylglycerol concentration, which was dramatically reduced in the 20400 fractions, was 195.1 nm and 347.8 nm, respectively. The lipid analysis showed that Sf>400 particles resembled the composition of natural chylomicrons and the 60<Sf<400 particles that of chylomicron remnants, being the range of particle size more homogeneous in the 60<Sf<400 fraction. The present is a reliable method for the preparation of CRLP, but also native chylomicron-like particles, in terms of lipid composition and particle size.

Formulation and Evaluation of Nanocrystals of a Lipid Lowering Agent

Atorvastatin calcium, the lipid lowering agent, is taken as a model drug characterized by poor water solubility and bioavailability. In this study an attempt was made for preparation of nanocrystals using high pressure homogenization. A number of stabilizers were included as well as polymers at different concentrations, and the formulations were homogenized for ten cycles at a pressure of 1000 ...

Preparation, statistical optimization and in vitro characterization of solid lipid nanoparticles as a potential vehicle for transdermal delivery of tramadol hydrochloride as a hydrophilic Compound

As encapsulation of hydrophilic drugs in the solid lipid nanoparticles (SLNs) is still a challenging issue, the aim of this study was to prepare SLNs containing tramadol hydrochloride as a hydrophilic compound. The SLNs were prepared using glycerol monostearate (GMS), soy lecithin and tween 80 by double emulsification-solvent evaporation technique. The nanoparticles were optimized through a cent...

Preparation and Evaluation of Acyclovir Liposomes by Two Techniques: A Comparative Study

The aim of this study was to prepare liposomes of acyclovir (ACY) by thin layer evaporation (TLE) and reverse phase evaporation (REV) methods. Twenty-seven batches of liposomes from each method were prepared using technique of three variables at three levels (33) factorial design. Drug/Lipid (molar ratio), hydration volume and hydration time were considered three independent variables in TLE me...
